Microbiota–gut–brain axis in neurodegenerative diseases: molecular mechanisms and therapeutic targets.
Molecular Biomedicine. 2025;6:64.
How the Gut Microbiota Communicates With the Brain👇
✅The microbiota–gut–brain axis is a complex, bidirectional communication system connecting the gut microbiota with the brain through the nervous, immune, and endocrine systems.
✅Gut microorganisms produce a wide range of bioactive molecules, including short-chain fatty acids (SCFAs), bile acids, neurotransmitter-related compounds, polyamines, tryptophan metabolites, and vitamins. These microbial products can influence intestinal and systemic physiology.
✅The vagus nerve and enteric nervous system (ENS) provide important neural routes through which signals from the gut can reach the brain. Gut-derived molecules can also influence the production and activity of neurotransmitters and neuroactive compounds.
✅The immune system forms another major communication pathway. Gut microbes interact with immune cells and intestinal tissues, influencing the production of cytokines and other inflammatory mediators that can affect brain function.
✅The gut also communicates with the brain through the hypothalamic–pituitary–adrenal (HPA) axis and other endocrine pathways. Intestinal epithelial cells can participate in sensing microbial signals and regulating neuroendocrine responses.
✅When the gut microbial community becomes disrupted, a condition known as dysbiosis, the production of beneficial metabolites may decrease while potentially harmful microbial products and inflammatory signals increase.
✅These changes can affect intestinal barrier function, immune regulation, neuroinflammation, and the integrity of the blood–brain barrier (BBB), potentially contributing to neurological dysfunction and disease progression.
