MBX Biosciences At MBX Biosciences, our mission is to help transform the lives of people living with obesity and rare endocrine disorders.

Using our peptide delivery platform, we are advancing a pipeline of novel Precision Endocrine Peptides (PEPs)™ designed to overcome key limitations of existing treatments and improve efficacy, tolerability, safety, and convenience. Our lead candidate, canvuparatide (MBX 2109) – a potential once-weekly parathyroid hormone (PTH) replacement therapy for the treatment of chronic hypoparathyroidism – is preparing for Phase 3 development. We are also advancing an obesity portfolio including MBX 4291, a GLP-1/GIP co-agonist prodrug in Phase 1 development, and MBX 5765, an amycretin prodrug in preclinical development. In addition, we are on track to nominate its GLP-1/GIP/GCG receptor prodrug candidate in Q3 2026, further expanding its obesity pipeline to potentially address the full spectrum of patient needs. Last month, MBX colleagues came together for Together Week, a chance to strengthen connections across teams and spend time with the patients and communities that inspire our work every day.

We kicked off the week by volunteering with a Life Science Cares Boston partner, The Food Project at Baker Bridge Farm, helping support its efforts to grow and distribute fresh, locally grown food throughout the region. We also had the privilege of hearing from a patient advocate living with hypoparathyroidism, whose story was a powerful reminder of why we do what we do.

This Together Week also marked an exciting milestone for MBX with the opening of our new office and laboratory facility in Burlington, Massachusetts. The expanded space and growing in-house capabilities will support our continued research efforts and help us build new connections within the Boston biotech community.

We're grateful to everyone who helped make the week so meaningful and look forward to building on this momentum in the months ahead.

It was a privilege to spend time with the hypoparathyroidism community at the 2026 International HypoPARA Conference in Phoenix last week.

The conference provided a valuable forum for members of our team to connect with patients, caregivers, advocates and health care professionals, hear their perspectives and better understand what matters most to this community.

Thank you to the HypoPARAthyroidism Association, Inc. for bringing the community together and creating an important space for connection, learning and support. We’re grateful to everyone who took the time to stop by our table and share their experiences.

At MBX, we are advancing precision peptide therapies that address significant unmet needs in endocrine and metabolic diseases.

At the forefront of this effort is our Discovery team. Working across disciplines, they collaborate closely with colleagues in CMC, Clinical, Regulatory, Commercial and Intellectual Property to identify and progress new candidates with the potential to become transformative therapies.

The recent nomination of our once-monthly GLP-1/GIP/glucagon agonist prodrug candidate is the latest example of that work in action. This development candidate expands our growing obesity portfolio and reflects the strength of our discovery engine.

John Lee, Director, Chemistry at MBX Biosciences, shared what it means to help build the next wave of MBX's pipeline:

"This team represents an important investment in MBX’s future. Together, we bring the expertise, scientific curiosity and shared commitment needed to advance promising science that could make a meaningful difference for people living with endocrine and metabolic disorders.”

We are proud to spotlight some of the scientists and innovators behind these discoveries and the work helping shape the future of our pipeline.

Today we are excited to announce that the first patient has been dosed in the pivotal Phase 3 oPTimize trial evaluating its once-weekly, investigational parathyroid hormone (PTH) replacement therapy for the treatment of adults with chronic hypoparathyroidism (HP).

The global registrational study will enroll approximately 160 patients and builds on positive Phase 2 Avail™ results and durable one-year data demonstrating sustained clinical benefit.

We look forward to advancing the oPTimize trial as we work to bring this potential therapy one step closer to people living with HP.

By 2035, 25% of the global population is projected to have obesity.

Meanwhile, many patients discontinue current treatments within 1–2 years – often driven by GI-related tolerability issues– highlighting the need for additional options.

Katherine Saunders, MD, FTOS, Co-Founder at FlyteHealth and Clinical Assistant Professor at Weill Cornell Medicine, shared these insights at our recent Obesity Day for investors.

At MBX Biosciences, we’re working to advance precision peptide therapies to help address these challenges for people living with obesity.
