This recent case report describes an exceptionally rare occurrence of advanced squamous cell lung cancer in a 25-year-old man with a history of short-term asbestos exposure and light smoking.

The disease was diagnosed at stage IVB with multiple bone and central nervous system metastases and progressed rapidly despite radiotherapy and first-line chemoimmunotherapy. The authors emphasize that lung cancer should be considered even in very young adults and that persistent respiratory symptoms warrant timely diagnostic evaluation.

A recent narrative review explores the efficacy of immune checkpoint inhibitors in rare oncogene-driven non-small cell lung cancer. BRAF-mutated NSCLC appears to derive the greatest benefit from immunotherapy, while ICI monotherapy shows more limited activity in MET exon 14, HER2, RET, and EGFR exon 20-altered tumors. Although chemoimmunotherapy may improve outcomes in selected patients, targeted therapies remain the preferred treatment strategy for most molecular subgroups.

This recent case report describes a rare pattern of dynamic HER2 discordance in breast cancer, with HER2 status changing over time and following systemic treatment. HER2 amplification was identified in the residual breast tumor after chemotherapy, while contralateral axillary metastases remained HER2-negative.

A recent narrative review summarizes the pathophysiology, clinical impact and current treatment strategies for cancer cachexia, a multifactorial syndrome associated with involuntary weight loss, skeletal muscle wasting, systemic inflammation and progressive functional impairment.

The authors emphasize that single-agent therapies provide limited benefits. Early, individualized multimodal management combining nutritional support, resistance and aerobic exercise, pharmacotherapy and emerging targeted therapies appears to offer the greatest potential to preserve muscle mass and physical function, improve treatment tolerance and maintain quality of life.

Nutritional strategies enriched with omega-3 fatty acids and high-protein formulas may support muscle preservation, while agents such as anamorelin provide partial symptomatic benefits. Emerging approaches targeting GDF-15, IL-6 and myostatin or activin signaling are promising, but require further clinical validation.

Prostate cancer in Poland, 2013–2023

A recent nationwide analysis shows that the age-standardized incidence of prostate cancer in Poland increased substantially between 2013 and 2023, while age-standardized mortality remained broadly stable. Despite a lower incidence than the EU-27 average, Poland continues to record considerably higher mortality, indicating a persistent gap in outcomes.

The study also documents the modernization of diagnostic pathways, including an approximately four-fold increase in MRI use, and a shift in first-line treatment: in 2023, surgery accounted for 53.0% of first treatments, compared with 40.8% for radiotherapy.

Nectins and nectin-like molecules are emerging as important players in cancer biology, immune regulation, and targeted therapy.

In this recent review, the authors summarize their expression, biological functions, prognostic relevance, and therapeutic implications in melanoma, lung cancer, and renal cell carcinoma. Nectin-4 and the CD155/TIGIT axis currently represent the most therapeutically actionable targets, while Nectin-1 has a dual role in melanoma as a metastasis suppressor and an entry receptor for HSV-based oncolytic therapy.

The review also discusses antibody-drug conjugates such as enfortumab vedotin, TIGIT-directed combination strategies, oncolytic virotherapy, and emerging CAR-T-cell approaches.

Interim data from the prospective UROCAPOL registry suggest that split-dose cisplatin may be underused in patients with metastatic urothelial carcinoma and moderate renal impairment.

Among patients treated with gemcitabine plus carboplatin who had evaluable creatinine clearance data, 71.0% had CrCl of 30–60 mL/min, the recognized renal eligibility window for split-dose cisplatin.

Compared with gemcitabine plus carboplatin, split-dose gemcitabine plus cisplatin was associated with a higher objective response rate to first-line chemotherapy, 91.7% versus 48.4%, and to subsequent avelumab maintenance, 66.7% versus 12.5%. Median avelumab treatment duration was also longer: 183.5 versus 85.0 days.

These interim observational findings should be interpreted cautiously and require prospective confirmation.

The place of atezolizumab in the treatment of elderly patients with non-small cell lung cancer — new opportunities and challenges

This recent review discusses the challenges of selecting first-line treatment for older patients with advanced NSCLC who are ineligible for platinum-based doublet chemotherapy.

Particular attention is given to the phase III IPSOS trial, in which atezolizumab monotherapy improved overall survival, nearly doubled the 2-year survival rate compared with single-agent chemotherapy (24.3% vs. 12.4%), and was associated with fewer grade 3–4 treatment-related adverse events (16% vs. 33%).

The article also presents a practical eligibility algorithm and highlights the importance of performance status, comorbidities, frailty, and comprehensive geriatric assessment when making treatment decisions.

This review highlights the role of DDR1 in colorectal cancer and its involvement in shaping the tumor microenvironment. Targeting DDR1 may offer a new strategy to overcome resistance to immune checkpoint inhibitors, particularly in pMMR/MSS colorectal cancer, and may represent a potential direction for future treatment development.
