WEIGHT LOSS PEPTIDES: Three Generations. One Goal.
Weight loss injectable medications have advanced rapidly across three distinct generations, each targeting more hormonal pathways than the last. Semaglutide, a GLP-1 single agonist, was the breakthrough — delivering approximately 14.9% body weight loss over 68 weeks and remaining the only option with proven cardiovascular benefits. Tirzepatide added a second receptor, GIP, achieving up to 22.5% weight loss and outperforming semaglutide by 47% in a direct head-to-head trial. Retatrutide, currently in Phase 3 trials, activates three receptors — GLP-1, GIP, and glucagon — producing up to 28.7% body weight loss in the TRIUMPH-4 trial, alongside an unprecedented 86% reduction in liver fat at 48 weeks. Each generation brings stronger efficacy but also distinct safety signals: pancreatitis risk with semaglutide, gallbladder events with tirzepatide, and dysesthesia — an abnormal sensation of tingling or numbness — emerging with retatrutide.
