NAD⁺: What the Evidence Actually Shows (sorry for everyone who thinks that he/she/it will get younger by using NAD+)
NR and NMN reliably increase NAD⁺ biomarkers — in some studies, blood NAD⁺ rises by around 50%.
But higher NAD⁺ does not currently mean proven better health.
A 2026 systematic review covering 33 human studies, including 28 randomized trials, found that while NR and NMN consistently raise NAD-related biomarkers, actual health benefits remain inconsistent or often absent — including effects on metabolism, cardiovascular function, physical performance, and other healthspan outcomes.
Bottom line: Higher NAD⁺ is proven. A meaningful anti-aging or health benefit in healthy humans is not - even though celebrities try to sell it that way. It’s just not the reality.
⚠️Side-Effects: At doses around 300 mg/day, no consistent serious adverse effects have emerged. Reported effects can include mild nausea, stomach/GI discomfort, headache, fatigue, or flushing, but these have not consistently occurred more often than with placebo.
The bigger uncertainty is long-term use. We do not yet have strong safety data for healthy people taking NR every day for many years. NR also increases breakdown products such as nicotinamide and methylated metabolites, but the clinical significance of this with long-term supplementation remains unclear.
