Allergens are usually harmless substances that, in some people, confuse the immune system, triggering allergic reactions. While research is still uncovering exactly how this happens, allergens appear to interact with the immune system in unique and sometimes exaggerated ways.

🫁🍽️ How Do Allergens Enter the Body?
Allergens can find their way in through everyday activities, such as:

• Breathing – airborne particles like pollen, dust mites, or pet dander
• Eating – foods such as milk, eggs, nuts, or seafood
• Stings or bites – venom from bees or wasps
• Medications – including antibiotics like penicillin or certain painkillers
• Skin contact – metals, dyes, or rubber (which can cause contact dermatitis without IgE involvement)

🔬 Main Types of Allergens

1️⃣ Inhaled Allergens
Pollen, dust mites, fungal spores, pet dander, insects

Trends in Immunology publishes an opinion article in which the authors review the potential role of trained immunity in protecting against respiratory infections and the development of mucosal bacterial vaccines based on trained immunity (TIbV).

The growing clinical burden of respiratory infections caused by viruses and/or bacteria makes it imperative to explore broader preventive approaches.

The article discusses how certain bacterial agents, such as the Bacillus Calmette–Guérin (BCG) vaccine and the polybacterial mucosal vaccine MV130, activate not only classical adaptive immunity but also “trained” innate functions, generating faster and more robust immune responses against multiple pathogens, including viruses. The concept of Trained Immunity-based Vaccines (TIbV) (https://www.frontiersin.org/journals/immunology/articles/10.3389/fimmu.2018.02936/full)applied to the respiratory mucosa is proposed as an effective strategy for respiratory tract protection.

The development of TIbV bacterial platforms for mucosal routes offers broad protection against respiratory infections. This approach could complement traditional vaccines by expanding the spectrum of prevention and modulating less-explored innate immune mechanisms. The article also addresses mechanisms of action, emerging clinical evidence, and discusses the regulatory and developmental challenges associated with this strategy.

The article published in the Journal of Fungi (https://www.mdpi.com/2309-608X/11/3/181) presents an extensive analysis of the polymerized extracts (allergoids) of Alternaria alternata, a highly relevant fungus in respiratory allergic sensitizations.

In the study, conducted at the R&D Department of Inmunotek, the physicochemical properties (SDS-PAGE, LC-MS/MS, NMR), proteolytic activity, IgE/IgG binding capacity, and immunogenicity in mice were evaluated. The results confirmed a significant reduction in IgE-binding capacity (i.e., a lower risk of allergic reactivity) while maintaining immunogenic capacity, generating a robust IgG response. Furthermore, when mixed with grass pollen extracts (Phleum pratense), the Alternaria allergoids improved the stability of the formulation compared to the unmodified native allergen.

The results of the study represent a significant advance in immunotherapy for polysensitized (polyallergic) patients allergic to fungi, pollens, or other allergens, as they open new avenues for safer, more stable, and more effective formulations.

The journal Frontiers in Allergy(https://www.frontiersin.org/journals/allergy/articles/10.3389/falgy.2025.1557650/full) published a study on the effect of glutaraldehyde polymerization of mite extracts on their proteolytic activity.

This study, conducted at INMUNOTEK, addresses a key issue in allergen immunotherapy (AIT) for polysensitized patients: the instability of allergen extract mixtures, particularly due to the high proteolytic activity of house dust mites (HDM). Using glutaraldehyde polymerization, the researchers evaluated whether this activity could be reduced in formulations combined with grass pollen allergens. Stability and antigenic activity were analyzed over 24 months, along with immunogenicity in a murine model.

The results showed a significant reduction in HDM proteolytic activity (up to 97.7%), which protected the grass allergens from degradation. Protein levels and antigenic activity remained stable throughout the study period. In mice, immune responses (IgG, T cell proliferation, and IL-10) were similar with or without the polymerized mite combination, demonstrating that immune efficacy was not compromised. Thus, polymerized extracts represent an effective and stable option for treating patients allergic to multiple allergens.

The journal Immunity (https://www.cell.com/immunity/fulltext/S1074-7613(24)00577-6) published a study on the effect of gut bacteria translocation on the induction of trained immunity in the bone marrow.

When the intestinal barrier is weakened, bacteria that normally live harmlessly in the gut can pass into the bloodstream, triggering inflammation. In this study, researchers gave mice a substance (DSS) that makes the gut more “leaky” and found that a common gut bacterium, Enterococcus faecalis, reached the bone marrow. There, it activated the immune system in a way that made it stronger and more responsive to future infections — a process known as “trained immunity.”

This effect was especially linked to a molecule called Mincle, which helps the immune system recognize bacteria. Mice that lacked Mincle didn’t show the same immune training and had less damage from the gut treatment. These findings suggest that signals from gut bacteria can “train” the immune system at its root — the bone marrow — and that this process could impact diseases linked to a leaky gut.
