RNA QC is just step one. Here are the 3 most common RNA extraction mistakes students make after that:
❌ Freeze-thaw cycles — degrade RNA silently. Always aliquot.
❌ RNase contamination — comes from your hands, not just the bench.
❌ Room temp delay — even 10 minutes after extraction matters.

🧬 DNA tells you what COULD happen. mRNA tells you what IS happening right now.

That is why RT-PCR measures mRNA — not DNA. And that is what your Ct value represents: how much of a specific gene is being expressed in your sample at the time of collection.

Lower Ct = higher expression. Higher Ct = lower expression. Simple concept, enormous implications for your thesis.
