🧠✨ Botox & Mood: More Than Wrinkle Relaxation?

Most people know botulinum toxin for softening expression lines—but research has explored an intriguing potential effect beyond aesthetics: improvement in depressive symptoms after treatment of the glabellar (frown) area.

A 2022 systematic review and meta-analysis combining 9 prospective studies, including 5 randomized controlled trials, found a significant reduction in depression scores following a single glabellar treatment with onabotulinumtoxinA. Improvements were generally evaluated around 6 weeks after treatment, with an overall large effect reported across the prospective studies.

Another meta-analysis of double-blind randomized trials also found onabotulinumtoxinA superior to placebo for unipolar major depression, with some analyses suggesting a greater effect in women.

💡 Why might the frown area matter? One proposed explanation is the facial feedback hypothesis: reducing persistent contraction of muscles associated with negative facial expressions may alter the feedback signals between the face and brain involved in emotional processing.

Important: Botulinum toxin is not FDA-approved as a treatment for depression, and these findings do not mean Botox should replace psychotherapy, medication, or psychiatric care. They do, however, represent an interesting and growing area of research into the connection between facial expression, the nervous system, and mood.

Studies referenced:
