New ASCO Living Guideline Update

Updated recommendations now include repotrectinib alongside entrectinib and larotrectinib for NTRK fusion-positive advanced solid tumors.

Key insights from the TRIDENT-1 trial for repotrectinib:

🔹 59% confirmed ORR in TKI-naïve patients (16% complete response)
High intracranial activity in patients with baseline CNS lesions
Distinct safety profile: Dizziness and dysgeusia were the most common TRAEs
🔹 Treatment choice should consider CNS involvement, patient comorbidities, and distinct toxicity profiles across available TKIs.
