Children's Cancer Therapy Development Institute cc-TDI's mission is to make all childhood cancers universally survivable regardless of diagnosis.

The Children’s Cancer Therapy Development Institute was created with one aim: to make childhood cancer universally survivable, regardless of diagnosis. For many forms of childhood cancer, we’ve seen little to no advancement in treatment options for over thirty years. To overcome this stalemate, cc-TDI is embarking upon a bold scientific course of discovery to create more effective clinical trials

for those childhood cancers that are most in need of treatments to save kids’ lives. Our research team is exploring and testing state-of-the-art treatment options for the most urgent issues facing children with cancer. By proving these methods in the laboratory, we can deliver smart, hope-filled discoveries to be prioritized in clinical trials for kids. Working in our own freestanding research institution allows us to act swiftly and nimbly to achieve results—bringing cost control, speed, purpose, and focus to translate effective treatments into the clinic. To get there, we perform basic science and translational research in our Innosphere lab at the base of the Rocky Mountains. Our mission is to translate scientific discovery into clinical trials by understanding and proving new disease-specific treatment options for children with cancer.

In our R&D and biopharma ecosystem of cc-TDI and , we congratulate Xiaolei Lian on his study of a tool ADC targeting the IL-13 receptor in the childhood brain tumor, DIPG. As mentioned in the paper, a different and in-licensed antibody is being pursued via the Artisan pipeline, but Xiaolei's paper gives proof-of-concept for the target (IL-13 receptor) and the payload type (a topoisomerase inhibitor). This therapeutic approach was born in an international collaboration we coordinated from the Children's Oncology Group brain tumor committee (a massive effort described in a Scientific American article, https://www.scientificamerican.com/article/scientists-tackle-lethal-childhood-brain-cancer).

In Xiaolei's newly-published studies, cc-TDI lab team members include Samuel, Victoria, Shefali, Guak-Kim and Noah, as well as interns Paige, Kyle and Emily. Collaborators included Drs. Hoshino, Liu and Lian. Artisan CSO, Dr. Tony Haight is also a key contributor and co-author. This work was funded by The Lyla Nsouli Foundation for Children's Brain Cancer Research, IronMattLarson, Hero Up Half Marathon, and young Ryan McGuire in affiliation with . Happy Birthday, Megan. We miss you. Charles & Team

💛 A message from the Bugg Family 💛

Today, Megan should have been celebrating her 25th birthday. We lost our precious Megan to childhood cancer on March 9, 2022. You would think that after four years, these birthday posts would get easier, but losing a child never really gets easier.

As the years pass, normal days become more tolerable, but the special occasions like her birthday bring everything back again. We miss our girl more than words can express. We miss her resilient human spirit, her kind heart, her amazing wisdom, and especially that beautiful contagious smile that could bring hope to the darkest of days.

On days like this, our family does its best to live life as Megan would have wanted: with optimism, hope, humility, compassion and happiness. In Megan's honor, we will continue fighting for her cause: to end the nightmare of childhood cancer.

No child should experience the physical and mental anguish of childhood cancer; and no parent should ever lose a child to this evil disease. Rest in peace Megan. I hope you take comfort in knowing that you made the world a better place. We will never stop loving you. P.S...Give Grandpa Bugg a hug for us.

In honor of the children, teens and young adults with the "rare" form of rhabdomyosarcoma harboring a PAX7::FOXO1 cancer protein (PAX-7, that is!), we have created and are now sharing with the scientific community a specialized genetic strain for studying this cancer's origins and mechanisms of progression.

The very international collaborative team included Roger Askew and Jonathan Gauntlett (Australia); Katherine Brechun and Harald Kranz (Germany); Jess Tate and Shireen Elhabian (Univ of Utah); and the cc-TDI Team consisting of Ellen Wang, Cassie Badger and Gillian St John (interns) as well as Naren Bharathy, Nicholas Salters and Samuel Rasmussen.

We are grateful for funding by the Megan’s Mission Foundation, the Pax7::Foxo1 Mouse International Crowdfunding campaign, and a very generous anonymous donor..

Congratulations to .kashyap on her publication of the Genomic Landscape of Myoepithelial Cancers. This study on a rare EWSR1-fusion cancer was inspired by Sebastián and supported by and . This true international collaboration including Paul Huang, Kwin Thway, Robin Jones and Yuen Bun Tam at ICR London; Sonja Chen, Ryan Roberts, Lindsay Ryan and Melissa Sammons at Nationwide Children's in Columbus; Morgan Darrow at UC-Davis; and Melvin Lathara and Ganapati Srinivasa at Datma. cc-TDI contributors include the gifted Hollis Wright, JiHyeon Byun (intern), Geoffrey Allintoff (intern) and Anandhi Mandi (research volunteer). The next step? Our EWSR1::KLF15 conditional genetic model (stay tuned for an update on this model!)
