Now enrolling! The Alliance A032103 (GAIN) trial is a phase III study evaluating whether adding gemcitabine, a well-established chemotherapeutic agent, to BCG can reduce high-grade recurrence rates in patients with NMIBC who have experienced recurrence after BCG.

New findings of an Alliance Foundation Trials study show combining immunotherapy and chemotherapy before surgery improves outcomes for some patients with locally advanced non-small cell lung cancer, according to the results of AFT-46/CHIO3 published in Lung Cancer.

“These results are exciting because they give us a new, successful blueprint for treating stage III lung cancer that has traditionally been difficult to manage,” said Linda W. Martin, MD, MPH, lead investigator, primary author of the study and a Professor of Thoracic Surgery at the University of Virginia. “This study shows that combining chemotherapy with immunotherapy can help more patients with lung cancer involving mediastinal nodes undergo complete cancer resection, improving their long term health.”

“To our knowledge, this is the first trial specifically focused lymph node clearance in this population, and the results look very promising,” said David Kozono, MD, PhD, Director of the Alliance Foundation, and Director of Thoracic Radiation Oncology at Mass General Brigham Cancer Center. “The high rates of lymph node clearance and successful surgical resection confirm this approach as an important option for patients with this stage of lung cancer.”

Preserving Cognitive Function in Advanced Prostate Cancer: New Data from AFT-47

For patients living with advanced prostate cancer, clinical efficacy is only part of the equation. Because many men will live years or decades after their initial treatment, the impact of therapies on daily functioning, memory, and overall quality of life is a critical factor in shared decision-making.

New data from the Alliance Foundation Trials (presented at ) sheds light on an important differentiator between two widely used androgen receptor inhibitors. The phase II ARACOG (AFT-47) trial directly evaluated the cognitive effects of darolutamide versus enzalutamide in patients with advanced prostate cancer.

Key Findings over 24 Weeks:
• Darolutamide: Patients experienced a 15.8% median decline in cognitive function.
• Enzalutamide: Patients experienced a 36.1% median decline in cognitive function.

While both agents are highly effective at controlling disease progression, the data demonstrates that darolutamide was associated with significantly less cognitive decline—more than halving the median decline seen with enzalutamide over the 24-week period.
