A study led by Mount Sinai researchers found that although the overall number of radiation oncology clinics in the United States has appeared stable in recent years, many individual treatment centers have quietly closed while others have opened — and the closures have disproportionately affected rural and freestanding practices.

Published in the International Journal of Radiation Oncology, Biology, Physics, the study found that freestanding practices were 56 percent more likely to close than hospital-affiliated sites, while rural practices also faced significantly greater risk.

"Understanding which communities are most vulnerable can help policymakers and health systems develop strategies to preserve access before these gaps become even larger," said senior author Kunal Sindhu, MD.

A new Mount Sinai study has found that patients with head and neck cancer who received chemotherapy in addition to immunotherapy before surgery had significantly greater tumor reduction at the time of operation than patients who received immunotherapy alone.

The study used real-world data to compare the two approaches before surgery for head and neck cancer. Researchers reviewed records of 86 adults treated at the Mount Sinai Health System over a two-year period. Of patients who received both chemotherapy and immunotherapy, roughly two-thirds had either no cancer or a very small amount of cancer remaining at the time of surgery.

“In this study, we found that giving a combination of chemotherapy with immunotherapy before surgery led to much greater destruction of the tumor than immunotherapy alone,” said Scott Roof, MD, MS.

Researchers at Mount Sinai have identified a role for the youth-associated protein TIMP2 in supporting the healthy function of microglia, the brain’s resident immune cells.

Published in Nature Portfolio, the study found that loss of TIMP2 caused microglia to develop several features associated with aging and neurodegeneration. Conversely, restoring TIMP2 in the blood of aged mice improved the ability of microglia to clear debris and reduced molecular markers associated with inflammation and other maladaptive states.

Aging is the strongest known risk factor for Alzheimer's disease and other neurodegenerative disorders, and microglia play an essential role in maintaining brain health. The findings provide new insight into how youth-associated factors may influence the aging brain and suggest that TIMP2 may help maintain healthy immune function in the brain as organisms age.

“I’m excited to take on the role of chair of the CV Section and build on the strong foundation that leaders before me have developed. We will continue to engage trainees and international societies, and to integrate artificial intelligence into cerebrovascular neurosurgery so we continue to further our mission of improving research and patient care,” said Dr. Kellner.

In his role, Dr. Kellner "will focus on enhancing access to research resources so that members of the CV section can access data that other members have developed and accumulated in clinical trials, and make that accessible and easily analyzable."

Researchers at Mount Sinai have found that exposure to per- and polyfluoroalkyl substances (PFAS), commonly known as "forever chemicals," during pregnancy and early life is associated with increased intestinal inflammation during childhood.

Published in Clinical Gastroenterology and Hepatology, the study measured PFAS concentrations in maternal blood collected during pregnancy, umbilical cord blood, and newborn dried blood spots, then followed children for up to 11 years. Higher PFAS mixture levels were associated with higher f***l calprotectin levels, a sensitive biomarker of intestinal inflammation, later in childhood. Both legacy PFAS compounds and newer replacement PFAS were linked to these findings, suggesting a broad range of these chemicals may influence children's gut health.

The findings provide new evidence that environmental exposures during critical stages of development may influence long-term intestinal health and future inflammatory bowel disease (IBD) risk.

Our researchers at the Icahn School of Medicine at Mount Sinai are among the Milestone 2 Awardee teams in XPRIZE Healthspan, a $101 million global competition that challenges researchers to develop new approaches that help people live healthier, longer lives.

XPRIZE Healthspan is a 7-year global competition incentivizing teams to develop and test therapeutics that restore muscle, cognition, and immune function in adults aged 50 to 90 by a minimum of 10 years, with an ambitious goal of 20 years, to extend healthy life within one year or less of treatment.

Single-cell "atlases" increasingly used to map the human body and train artificial intelligence models may not represent the world's populations fairly, according to a study led by Dr. Kuan-lin Huang, published in Cell Genomics.

The team reviewed more than 13,500 samples from three major single-cell resources — the Human Cell Atlas, the Human Tumor Atlas Network, and the PsychAD Consortium — and found that people of European ancestry were consistently overrepresented. At the same time, Asian and Latino individuals were underrepresented. A large share of samples also lacked any record of ancestry at all.

The concern, the authors say, is one of fairness and reliability: if future research, AI tools, biomarkers, or treatments are built on datasets that do not represent everyone, the benefits of those advances may not reach everyone equally. The team also provides a practical, field-specific checklist that research groups can use when designing future studies, covering recruitment, demographic record-keeping, sample balancing, and AI model performance across ancestry and s*x groups.

Researchers at Mount Sinai have identified immune system changes that can be detected up to 10 years before inflammatory bowel disease (IBD) develops, offering new insight into how the disease begins.

Published in Gut, the study is one of the most comprehensive investigations to date of the preclinical phase of IBD. Using antibody profiling technology, researchers analyzed nearly 2,000 blood samples collected over about a decade from people who later developed Crohn's disease or ulcerative colitis.

They identified unique antibody signatures against viruses and bacteria — particularly Epstein-Barr virus and bacterial flagellins — that were present years before diagnosis, suggesting that abnormal immune activity begins long before the disease becomes clinically apparent.

The findings could eventually help identify people at increased risk for IBD, particularly relatives of patients with the disease, and guide future prevention strategies.
