In this week’s Featured Image, Coulombe-Leveque et al. depict key pathophysiological changes in PTSD and nociplastic pain that form the basis of a proposed PTSD-inspired pathophysiological and conceptual model for nociplastic pain, wherein the disproportionate threat response is driven by increased activity in the amygdala, enhanced by increased excitatory drive from the locus-coeruleus, with dysfunctional modulation by the vmPFC and hippocampus.
