Locally manufactured versus commercial CAR T in large B-cell lymphoma.
As CAR T-cell therapy expands globally, questions remain regarding the comparative performance of locally manufactured and commercial products. A new multicenter propensity score-matched analysis examines outcomes in patients with large B-cell lymphoma, addressing a key issue in accessibility, scalability, and real-world cellular therapy practice.

Predicting thrombo-ischemic risk in acute promyelocytic leukemia.

Despite major therapeutic advances, thrombo-ischemic complications remain a serious concern in acute promyelocytic leukemia. In the PETHEMA/PALG study, investigators developed a scoring system aimed at identifying patients at higher risk of life-threatening thrombo-ischemic events, addressing an important challenge in risk stratification and supportive care. Targeting RNA-binding proteins in leukemia.
RNA-binding proteins are emerging as promising therapeutic targets in hematologic malignancies. The RNA-binding protein IGF2BP3 is an oncofetal protein over-expressed in B-cell acute lymphoblastic leukemia and is critical for leukemogenesis in experimental models. A new study investigates a small molecule inhibitor of IGF2BP3, exploring how disruption of RNA regulatory networks may affect leukemic cell survival and open new avenues for targeted therapy in leukemia. Idiopathic multicentric Castleman disease (iMCD) encompasses biologically distinct subtypes with variable inflammatory profiles. The two major clinical subtypes are idiopathic plasmacytic lymphadenopathy (IPL) and iMCD with thrombocytopenia, anasarca, fever, renal dysfunction/reticulin fibrosis, and organomegaly (TAFRO). A new study examines interleukin-6 production in IPL and TAFRO subtypes, exploring differences in cytokine regulation and their implications for disease classification and therapeutic strategies.

Mantle cell lymphoma (MCL) remains a challenging and generally incurable disease. Treatment outcomes in MCL have evolved substantially over the past decades. A pooled analysis of six randomized phase III trials (1996–2020) examines survival trends in patients ≤65 years with advanced-stage disease, addressing how therapeutic advances have reshaped long-term outcomes in this population. Despite standard treatment with hypomethylating agents, the prognosis of patients with higher-risk myelodysplastic syndrome (MDS) remains poor and treatment strategies must evolve. A new multicenter, randomized controlled trial compares decitabine plus all-trans retinoic acid with decitabine monotherapy, examining whether combination approaches may refine outcomes in this high-risk patient population. Donor selection in allogeneic transplantation: does age matter? Donor selection remains a cornerstone of allogeneic hematopoietic stem cell transplantation, yet priorities may shift across different patient populations. A new study explores age-related differences in donor selection strategies, examining how clinical decision-making may vary and what this means for optimizing transplant outcomes.

The clearance of red blood cells is a tightly controlled process central to hematologic homeostasis. This study investigates the role of ALX/FPR2 signaling in splenic erythrophagocytosis, exploring how immune pathways regulate red cell turnover and contribute to physiological and pathological processes.
