Nasosorption® used to measure omalizumab in nasal mucosal lining fluid

A Genentech study used Nasosorption® FX·i to assess whether omalizumab could be detected in nasal mucosal lining fluid following treatment.

Omalizumab is an anti-IgE monoclonal antibody used in allergic disease. Measuring drug levels in blood does not necessarily show how much treatment reaches the airway surface, where local disease activity occurs.

The study collected matched serum and Nasosorption® samples from eight patients over a complete dosing cycle.

The researchers found that:

• omalizumab was detectable in nasal mucosal lining fluid following treatment;
• the median nasal-fluid-to-serum partitioning ratio was 0.52%;
• Nasosorption® provided a minimally invasive way to assess drug distribution into the upper airway; and
• local sampling could support dose selection for treatments targeting airway disease.

This is a useful example of how Nasosorption® can support clinical pharmacology by measuring drug exposure directly at the airway surface, rather than relying only on blood measurements.

Nasosorption® used to measure treatment activity directly in the upper airway

A Phase 1 study from Genentech used Nasosorption® to measure active tryptase in nasal mucosal lining fluid following treatment with MTPS9579A, an investigational anti-tryptase antibody.

The study included 106 healthy volunteers and found:

• a rapid reduction in active tryptase following treatment;
• a clear dose-dependent response in Nasosorption® samples;
• evidence of pharmacodynamic activity directly within the upper airway; and
• a PK/PD relationship that could help inform future clinical trials.

The researchers concluded that the investigational antibody inhibited tryptase activity in the upper airway. This provides a useful example of how minimally invasive mucosal sampling can support local biomarker measurement, dose-response assessment and early clinical-trial decision-making.

Genentech research using Nasosorption® samples

Researchers at Genentech evaluated whether urea could help correct for variable dilution in nasal mucosal lining fluid collected using Nasosorption®.

Nasosorption® is used to collect nasal mucosal lining fluid for biomarker and drug-level analysis. Because absorptive sampling can recover different amounts of fluid, reliable methods of accounting for dilution are important when comparing results between samples.

The study assessed matched serum and Nasosorption® samples from healthy donors, biologic-treated participants and people enrolled in a severe-asthma clinical trial.

The researchers found that the Nasosorption-to-serum urea ratio was stable across participant groups and over repeated sampling. They concluded that it could be useful for adjusting for variable analyte dilution across Nasosorption® samples.

This is a useful example of how minimally invasive airway sampling can be combined with robust analytical methods to support respiratory biomarker and clinical pharmacology research.

Nasosorption® FX·i “SAM strips” used for long-term home sampling in UK families

A study published in Scientific Reports, part of the Nature Portfolio, assessed minimally invasive self-sampling at home for longitudinal monitoring of the oral, nasal and hand microbiota within UK families.

Participants collected nasal lining fluid at home every two weeks for six months using Synthetic Absorptive Matrix strips supplied by Hunt Developments. The publication refers to these as “SAM strips” and identifies Hunt Developments as the supplier, with 3.0 mm, 4.5 mm and 7.0 mm sizes selected according to participant age.

Across the study:

• 1,557 nasal lining fluid samples were returned;
• compliance exceeded 80% across sample types and age groups;
• families described the wider sampling approach as fast, easy and painless; and
• the researchers concluded that at-home multi-site sampling could support larger longitudinal studies of respiratory microorganisms.

The devices described as “SAM strips” are part of the proprietary Nasosorption® FX·i platform, designed and manufactured exclusively by Hunt Developments (UK) Limited under the Mucosal Diagnostics™ brand.

Nasosorption® FX·i in COVID-19 mucosal immunity research

A 2025 study published in The Journal of Infectious Diseases evaluated systemic and mucosal immune responses following a bivalent COVID-19 booster.

Nasal samples were collected using Nasosorption® FX·i and FLOQSwab, while saliva was collected using Oracol.

Compared with FLOQSwab samples, Nasosorption® samples contained:

• 1.2–3.4 times more total nasal IgG
• 1.3–2.7 times more total nasal IgA

Across the three mucosal collection approaches, the highest measured antibody levels were found in nasal samples collected using Nasosorption®.

Normalised virus-specific antibody concentrations were similar between the two nasal collection methods, but the study demonstrates the strong recovery of nasal antibodies achieved using Nasosorption® FX·i.

What is Nasosorption™ and how is it used in respiratory research?

Nasosorption™ refers to both a nasal swab device and a validated sampling method used to collect mucosal lining fluid from the surface of the nasal airway.

The device contains a synthetic absorptive matrix (SAM) that gently absorbs fluid from the nasal mucosa. It’s non-invasive and ideal for biomarker analysis, including cytokines, chemokines, proteins, and viral RNA, in clinical trials and translational research.

Imperial College London has published a peer-reviewed video protocol in the Journal of Visualized Experiments (JoVE), demonstrating the full sampling and processing workflow using the Nasosorption FX·i device.
