GIMM researchers, led by Edgar Gomes, have published a new study uncovering a molecular brake that controls muscle architecture.

The study shows how a specialised actin network, involving Arp2/3 complexes containing the Arpc5 subunit, restrains the growth of T-tubules, the internal membrane system that enables muscle fibres to contract in a coordinated way. Without this brake, T-tubules become disorganised and muscle contraction becomes less cordinated.

These findings offer a new perspective on how muscle architecture is built and maintained, opening new avenues to understand what goes wrong in muscle disorders such as congenital myopathies and muscular dystrophies.

GIMM researchers André Ventura-Gomes, Rui Sousa-Luís and Maria Carmo-Fonseca have published a new review in Current Opinion in Structural Biology, bringing together the latest advances in our understanding of nuclear speckles, structures inside the cell nucleus once thought to act mainly as storage sites.

The review highlights a far more active role for these structures in coordinating RNA processing, from splicing and quality control to the export of mature RNA. It also explores how the position of genes relative to nuclear speckles can influence how efficiently RNA is processed.

These findings are reshaping our understanding of how the cell nucleus is organised and raising new questions about the relationship between nuclear architecture, cell function and disease.
